最近開始要做wound healing assay, 可是在實驗設計上遇到幾個問題
想請教一下大家
我先把我的實驗條件跟使用細胞講一下
我的細胞是293 overexpress A gene(也就是我要看的gene) 還有vector
這些都是 stable clone
以下這是我的protocol
我是用6-well plate 來做實驗,
前一天先seeding 1.6*10^6/ well, incubate 16-18hr後, 先用2ml 1x PBS wash 一次,
之後加入low-serum mediun(FBS conc: 0.5%) starve 24 hr, 用200ul tip 劃線後,
wash之後再加入我的condition medium
然後我有幾個問題想請教大家
1. 昨天種了1.6*10^6/ well之後,
今天早上來看,發現細胞的貼附情況並不是很好
(而且輕輕搖晃plate ,細胞會有飄浮起來的現象),
雖然說293本身的貼附能力並沒有很好,
但我之前遇到這種情況都是293已經長滿,才會有這種漂浮的現象,
想請問大家如果有沒有遇過這種情況?
然後293 種在6 –well 裡面通常都是種多少的細胞數比較好呢?
2. 假設我把細胞種少一點(但一開始vector 跟a gene stable種的細胞數是一樣的), 等
它們長滿再開始做實驗,請問這樣會影響到實驗結果跟誤差嗎?
3. 如果我只是要看normal狀態下,這些stable clone的爬行能力(也就是說我的
condition medium裡沒有加其他的東西)
請問FBS 濃度用多少才好?
